The 18-year-old has signed a four-year contract, with an option for a further 12 months, at Old Trafford having secured the move after returning from World Cup duty with England.
United had an initial bid of £27m for Shaw turned down at the end of last season but an improved offer sealed the deal.
The teenager, who made a single appearance in the World Cup, joins Ander Herrera in moving to Manchester; the midfielder is moving on a four-year contract from Athletic Bilbao and for a fee of around £28m.
Shaw said: "I am thrilled and excited to be joining Manchester United. I have been at Southampton since I was eight years old and I would like to thank them and the fans for everything they have done for me.
"I want to continue to progress my career and joining United is the ideal place for me to do that. I am looking forward to this new chapter in my life and also looking forward to learning from the world-class players and management at the club.”
